    <ns1:description language="de">Ziel dieser Studie war es, die Effekte von Eigelb und Eidotterplasma sowie von Katalase in verschiedenen TiefkÃ¼hlverdÃ¼nnern zur Samentiefgefrierung von Hunden zu untersuchen. In der Diplomarbeit von Burak und Papadopoulos (2019) wurden Samenproben von 17 gesunden RÃ¼den verschiedener Rassen gewonnen, tiefgefroren und nach dem Auftauen auf kinetische Parameter, Viability, Morphologie, Apoptose, Reactive oxygene species (ROS) und DNA SchÃ¤den untersucht. Im Rahmen der vorliegenden Studie wurden die gleichen Proben auf ihre BindungsfÃ¤higkeit an kanine Oozyten untersucht. Die fraktionierte Samengewinnung wurde bei jedem Hund einmal durchgefÃ¼hrt, nach einer QualitÃ¤tsÃ¼berprÃ¼fung wurden die Samenproben ohne Zentrifugation in drei Aliquots aufgeteilt. Die Aliquots wurden mit KÃ¼hlmedien verdÃ¼nnt (I, II, III), danach wurden die Proben eine Stunde bei +4 Â°C im Wasserbad Ã¤quilibriert und dann das TiefkÃ¼hlmedium dazu gemischt (1:1, EndverdÃ¼nnung 1:3). Die Probe A enthielt TRIS- Fructose-ZitronensÃ¤ure VerdÃ¼nner mit Eidotter, Glyzerol und Equex STM Paste. Die Probe B enthielt die gleiche Komponente, aber statt Eidotter wurde Eigelbplasma verwendet. Bei der Probe C wurde ein TRIS-Lecithin VerdÃ¼nner mit Eidotterplasma, Glyzerol und Equex STM Paste verwendet. Das jeweils zweite Aliquot von Probe B und C wurde zusÃ¤tzlich mit 300 I.U./mL Katalase angereichert (Bcat und Ccat). Nach AbfÃ¼llung in 0,5 ml Pailletten, wurden die Proben mit einem Computer assoziierten Einfrierautomat gefroren (SchÃ¤fer-Somi et al. 2006) und bis zur Analyse im flÃ¼ssigen Stickstoff gelagert. Nach dem Auftauen wurde eine Samenprobe mittels Computer Assisted Sperm Analysis (CASA) untersucht sowie eine weitere mittels Durchflusszytometrie auf DNA SchÃ¤den (Sperm Chromatin Structure Assay; SCSA), reactive oxygen species (ROS) und Apoptose. Mit dem Samen einer weiteren Paillette wurde ein Zona Binding Assay (ZBA) durchgefÃ¼hrt. Die Proben mit Eidotter und Equex STM Paste zeigten eine bessere progressive MotilitÃ¤t (P), den hÃ¶chsten Prozentsatz an intakten Zellen und den geringsten Prozentanteil an AkrosomschÃ¤den und signifikant weniger ROS (p&lt;0.05 bzw p&lt;0.01). Die Proben mit Katalase und Lecithin und Equex STM Paste waren nicht besser, sie enthielten die meisten toten Zellen. Ersatz von Eidotter durch Eidotterplasma erhÃ¶hte die Zahl der Zellen mit spÃ¤ter Apoptose und die ROS signifikant (Burak und Papadopoulos 2019). In unser Studie zeigten die Proben mit verschiedenen VerdÃ¼nnungsmedien keinen signifikanten Unterschied hinsichtlich der BindungsfÃ¤higkeit. Die Schlussfolgerung ist, dass ein Zwei-PhasenverdÃ¼nner mit TRIS-EY, Glycerol und Equex STM Paste (A) besser hinsichtlich P, Viability, ROS und Apoptose war, als die anderen TiefkÃ¼hl-Medien. Die Ergebnisse von TRIS-EYP mit Glycerol und Equex STM Paste waren Ã¤hnlich, allerdings waren mehr ROS und apoptotische Zellen nachweisbar. Die in-vitro BindungskapazitÃ¤t und VCL waren nicht signifikant unterschiedlich zwischen den Gruppen. Anscheinend waren genÃ¼gend motile und Membran-intakte Zellen vorhanden, um die Bindung zu gewÃ¤hrleisten. Die FertilisationskapazitÃ¤t dieser VerdÃ¼nnungsmedien muss jedoch in-vivo untersucht werden.</ns1:description>
    <ns1:description language="en">The aim of this study was to assess the effects of egg yolk, egg yolk plasma and catalase in different freezing extenders on frozen-thawed semen in dogs. In the study of Burak und Papadopoulos (2019), semen samples (main fraction) were collected from 17 healthy dogs of different breeds. The samples were frozen, and after thawing, the quality of the samples was analyzed and parameters like kinetics, viability, morphology, apoptosis, reactive oxygen species (ROS) and DNA damage evaluated. In this study, the in-vitro binding ability of the previously mentioned samples was examined. Fractionated semen collection was done once from each of the dogs. After quality evaluation of the specimens, they were devided without centrifugation into three aliquots. The aliquots were diluted 1:1 with cooling extender (I, II, III), afterwards the samples were equalibrated for one hour in a water bath at+4 Â°C, then the freezing extender was added (1:1, final dilution rate 1:3). Sample A contained TRIS-fructose-citric acid extender with egg yolk, glycerol and Equex STM Paste. Sample B contained the same components, but egg yolk plasma was used, instead of egg yolk. In sample C, a TRIS âbased lecithin extender was used with egg yolk plasma, glycerol and Equex STM Paste. The second aliquots of sample B and C were enriched with 300 I.U./mL catalase (Bcat and Ccat). After filling in 0,5 ml straws the samples were frozen in a computer-assisted freezing automat (SchÃ¤fer-Somi et al 2006) and stored in liquid nitrogen until analysis. After thawing, a computer assisted sperm analyser (CASA) was used for quality assessment. Furthermore, flow cytometry was used for a sperm chromatin structure assay (SCSA) and examination for ROS and apoptosis. Finally, a Zona Binding Assay (ZBA) was done with semen from one more straw. The samples with egg yolk and Equex STM Paste showed better progressive motility (P), the highest percentage of intact cells and the lowest percentage of acrosome damage and significant less ROS (p&lt;0.05 and p&lt;0.01) than the other samples. The samples with catalase and lecithin were not better, the amount of dead cells were at maximum. The replacement of egg yolk with egg yolk plasma increased the number of cells with late apoptosis and the ROS significantly (Burak und Papadopoulos 2019). In our study the samples with different extenders showed no significant difference in their binding ability. The final conclusion is that a two-phase freezing medium with TRIS-EY, glycerol and Equex STM paste (A) is better with regards to P, viability, ROS and apotosis. The results of TRIS-EYP, glycerol and Equex STM Paste were similar, though more ROS and apototic cells were detected.. In-vitro binding ability and VCL did not differ significantly between groups. Apparently there were enough motile and membrane intact cells to achieve binding. The fertilizing capacity of the extenders should be examined in-vivo.</ns1:description>
