    <ns1:description language="de">Sehnenpathologien sind sowohl in der Humanmedizin, als auch in der VeterinÃ¤rmedizin Gegenstand vieler Forschungsprojekte, da der Heilungsprozess und die Genesung von Sehnengewebe langwierig ist. Zur Erforschung solcher Fragestellungen kommen immer hÃ¤ufiger Bioreaktoren zum Einsatz, deren Aufgabe es ist, Kulturbedingungen so naturgetreu wie mÃ¶glich nachzuahmen. Zu diesem Zweck wurden im Rahmen des Studienprojektes âEntwicklung eines 3D in vitro Sehnenmodellsâ - eine Machbarkeitsstudie Sehnenzellen in vitro kultiviert und in ein speziell fÃ¼r dieses Projekt designtes 3D Modell Ã¼bertragen. Ziel der hier vorgestellten Machbarkeitsstudie war zum einen die erfolgreiche Herstellung von Sehnenkonstrukten mit sehnenÃ¤hnlicher Morphologie, zum anderen die DurchfÃ¼hrbarkeit geeigneter Analyseverfahren, wie Histologie und qPCR, um die in vitro Versuche Ã¼berprÃ¼fen zu kÃ¶nnen zu testen. Es wurden fÃ¼nf verschiedene Versuche mit Hilfe des 3D Modells durchgefÃ¼hrt. Das Hauptaugenmerk der Versuche lag auf der Ermittlung der am besten geeigneten Kollagen-, Eisennanopartikel- und Zell-Konzentration, um die besten Bedingungen fÃ¼r die Nachahmung sehnenartigen Gewebes zu schaffen. Die besten Ergebnisse wurden bei einer Kollagenkonzentration von 5 mg/ml und 6,5 mg/ml, einer SPIO-Konzentration zwischen 0,005â0,0005 mg/ml und einer optimalen Zellzahl von 660â800 Zellen/Î¼l erzielt. Da das Wachstum einer Sehne in vivo von statischen und dynamischen Zugspannungen beeinflusst wird, wurden diese Faktoren im 3D Modell ebenfalls berÃ¼cksichtigt. Die statische Zugspannung wurde mit Hilfe von feststehenden Pins erzeugt. Die dynamische Zugspannung wurde mittels MagnetwÃ¼rfeln erzeugt, welche die mit Eisennanopartikeln beladenen Zellen in longitudinaler Richtung uniaxial dehnen sollten. Die dynamische Aktivierung der Sehnen- Organoide mit Hilfe von Magneten war nicht mÃ¶glich, da sie die Zellen zum Platzen brachten. Es wurde deshalb ausschlieÃlich mit der statischen Zugspannung weitergearbeitet. Die Auswertung erfolgte makroskopisch, mittels Histologie und qPCR. Die Sehnen-Organoide der 3D Modelle zeigten eine Genexpression, die mit einem Remodellierungsprozess in Verbindung gebracht werden kÃ¶nnten. Zusammenfassend demonstrierte die hier vorgestellte Machbarkeitsstudie, dass eine Anzucht von Sehnen-Organoiden mit sehnenÃ¤hnlicher Morphologie gelang und dass sie sich zur Analyse mittels Histologie und qPCR eignen.</ns1:description>
    <ns1:description language="en">Tendon pathologies are subject of many research projects in both human and veterinary medicine, because the healing process and recovery of tendon tissue is a long process. Bioreactors are increasingly being used to investigate such issues. Goal is to provide with unsatisfying outcome culture conditions closely mimicking the natural cell environment. For this purpose, tendon cells were cultivated in vitro and transferred into a 3D model specially designed for this project within the scope of this study âDevelopment of a 3D in vitro tendon modelâ â a feasibility study. The aim of the feasibility study presented here, was on the one hand, the successful production of tendon constructs with tendon-like morphology and, on the other hand, to test the feasibility of suitable analytical procedures, such as histology and qPCR performed on the developed tendon organoids. Five different experiments were performed using the 3D model. The main focus of the experiments was to determine the most suitable collagen, iron nanoparticle and cell concentration to create the best conditions for imitating tendon-like tissue. The best results were achieved with collagen concentrations between 5 mg/ml and 6.5 mg/ml, SPIO concentrations between 0.005-0.0005 mg/ml and an optimal cell count of 660-800 cells/Î¼l. Since the growth of a tendon in vivo is influenced by static and dynamic tensile stresses, these factors were also considered in the 3D model. The static tensile stress was generated using fixed pins. The dynamic tensile stress was generated by magnetic cubes which exerted uniaxial directed magnetic forces onto the cells loaded with iron nanoparticles to stretch the cells uniaxially in longitudinal direction. The dynamic activation of the tendon organoids with magnets and SPIO particles was not possible because they caused the cells to burst. Therefore, only the static tensile stress was successful. The resulting tendon organoids were evaluated macroscopically, histologicallly and by qPCR. The tendon organoids of the 3D models showed gene expression that could be indicative of a remodeling process. In summary, the feasibility study demonstrated that a cultivation of tendon organoids with tendon-like morphology was successful and that the tendon organoids can be analyzed by histology and qPCR.</ns1:description>
