<lom:langstring xml:lang="en">Equine coronaviruses (ECoV), which belong to the betacoronaviruses (ß-CoV), can cause fever, lethargy, anorexia and gastrointestinal disorders such as diarrhea and colic in horses. Diseased animals often show leukopenia and less frequently neurological symptoms, such as ataxia. As previous studies have focused on outbreaks and determination of seroprevalence, the aim of this thesis was to examine equine faecal samples from healthy animals for equine and bovine coronaviruses using molecular biology. As part of the present work, a duplex RT-qPCR was established in which ß-CoV-1-specific nucleic acids and the EGFP gene (enhanced green fluorescent protein), added as an internal control, can be detected simultaneously. Absolute quantification using a standard dilution series prepared in triplicates showed, that this duplex assay can reliably detect 10 copies of the virus genome per PCR reaction. Subsequently, 588 horse faeces samples from Austria were tested, which had previously been collected as part of a parasitological study. After the preparation of faecal supernatants, these were extracted in pools of three samples each and then the validated ß-CoV1-EGFP duplex RT-qPCR was performed. As no ß-CoV1-specific nucleic acids were detected in any of the faecal samples examined, it can be assumed that clinically healthy horses pose little risk of spreading the virus with regard to equine and bovine coronaviruses. However, due to the tendency of coronaviruses to mutate and the associated zoonotic potential, it is crucial to continue monitoring betacoronaviruses closely.</lom:langstring>
