This study aimed to further elucidate the role of vitamin D signaling in the development of left ventricular (LV) hypertrophy and dysfunction. To ablate the vitamin D receptor (VDR) specifically in cardiomyocytes, VDRfl\/fl mice were crossed with Mlcv2-Cre mice. To induce LV hypertrophy experimentally by increasing cardiac afterload, transverse aortic constriction (TAC) was employed. Sham or TAC surgery was performed in 4-month-old, male, wild-type, VDRfl\/fl, Mlcv2-Cre, and cardiomyocyte-specific VDR knockout (VDRCM-KO) mice. As expected, TAC induced profound LV hypertrophy and dysfunction, evidenced by echocardiography, aortic and cardiac catheterization, cardiac histology, and LV expression profiling 4 weeks post-surgery. Sham-operated mice showed no differences between genotypes. However, TAC VDRCM-KO mice, while having comparable cardiomyocyte size and LV fibrosis to TAC VDRfl\/fl controls, exhibited reduced fractional shortening and ejection fraction as measured by echocardiography. Spatial transcriptomics of heart cryosections revealed more pronounced pro-inflammatory and pro-fibrotic gene regulatory networks in the stressed cardiac tissue niches of TAC VDRCM-KO compared to VDRfl\/fl mice. Hence, our study supports the notion that vitamin D signaling in cardiomyocytes plays a protective role in the stressed heart."}]}],"bf:provisionActivity":[{"@type":"bf:Publication","bf:agent":[{"@type":"schema:Organization","schema:name":[{"@value":"MDPI"}]}]}],"dce:rights":[{"@language":"eng","@value":"© 2024 by the authors"}],"dce:subject":[{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Vitamin D"}]},{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Vitamin D Receptor"}]},{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Left Ventricular Hypertrophy"}]},{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Cardiomyocytes"}]},{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Spatial Transcriptomics"}]},{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Inflammation"}]},{"@type":"skos:Concept","skos:prefLabel":[{"@language":"eng","@value":"Fibrosis"}]}],"dce:title":[{"@type":"bf:Title","bf:mainTitle":[{"@language":"eng","@value":"Ablation of Vitamin D Signaling in Cardiomyocytes Leads to Functional Impairment and Stimulation of Pro-Inflammatory and Pro-Fibrotic Gene Regulatory Networks in a Left Ventricular Hypertrophy Model in Mice"}]}],"dcterms:accessRights":[{"@type":"skos:Concept","skos:exactMatch":["https:\/\/pid.phaidra.org\/vocabulary\/QW5R-NG4J"],"skos:prefLabel":[{"@language":"eng","@value":"open access"}]}],"dcterms:issued":["2024"],"dcterms:language":["eng"],"dcterms:type":[{"@type":"skos:Concept","skos:exactMatch":["https:\/\/pid.phaidra.org\/vocabulary\/69ZZ-2KGX"],"skos:prefLabel":[{"@language":"eng","@value":"Text"},{"@language":"deu","@value":"Text"},{"@language":"ita","@value":"Testo"}]}],"ebucore:filename":["ijms-25-05929.pdf"],"ebucore:hasMimeType":["application\/pdf"],"edm:hasType":[{"@type":"skos:Concept","skos:exactMatch":["https:\/\/pid.phaidra.org\/vocabulary\/VKA6-9XTY"],"skos:prefLabel":[{"@language":"eng","@value":"journal article"},{"@language":"deu","@value":"Wissenschaftlicher Artikel"},{"@language":"ita","@value":"Articolo di rivista"}]}],"edm:rights":["http:\/\/creativecommons.org\/licenses\/by\/4.0\/"],"oaire:version":[{"@type":"skos:Concept","skos:exactMatch":["https:\/\/pid.phaidra.org\/vocabulary\/PMR8-3C8D"],"skos:prefLabel":[{"@language":"eng","@value":"version of record (published version)"}]}],"rdam:P30004":[{"@type":"ids:doi","@value":"10.3390\/ijms25115929"}],"rdau:P60193":[{"@type":"schema:CreativeWork","bibo:issue":["11"],"bibo:volume":["25"],"dce:title":[{"@type":"bf:Title","bf:mainTitle":[{"@language":"eng","@value":"International Journal of Molecular Sciences"}]}],"dcterms:issued":["2024"],"ids:issn":["1422-0067"]}],"role:aut":[{"@type":"schema:Person","schema:affiliation":[{"@type":"schema:Organization","schema:name":[{"@value":"University of Veterinary Medicine Vienna"}],"skos:exactMatch":["https:\/\/ror.org\/01w6qp003"]}],"schema:familyName":[{"@value":"Zupcic"}],"schema:givenName":[{"@value":"Ana"}]},{"@type":"schema:Person","schema:affiliation":[{"@type":"schema:Organization","schema:name":[{"@value":"University of Veterinary Medicine Vienna"}],"skos:exactMatch":["https:\/\/ror.org\/01w6qp003"]}],"schema:familyName":[{"@value":"Latic"}],"schema:givenName":[{"@value":"Nejla"}],"skos:exactMatch":[{"@type":"ids:orcid","@value":"0000-0001-5635-5175"}]},{"@type":"schema:Person","schema:familyName":[{"@value":"Oubounyt"}],"schema:givenName":[{"@value":"Mhaned"}]},{"@type":"schema:Person","schema:affiliation":[{"@type":"schema:Organization","schema:name":[{"@value":"University of Veterinary Medicine Vienna"}],"skos:exactMatch":["https:\/\/ror.org\/01w6qp003"]}],"schema:familyName":[{"@value":"Ramesova"}],"schema:givenName":[{"@value":"Alice"}],"skos:exactMatch":[{"@type":"ids:orcid","@value":"0000-0001-7482-7818"}]},{"@type":"schema:Person","schema:familyName":[{"@value":"Carmeliet"}],"schema:givenName":[{"@value":"Geert"}]},{"@type":"schema:Person","schema:familyName":[{"@value":"Baumbach"}],"schema:givenName":[{"@value":"Jan"}]},{"@type":"schema:Person","schema:familyName":[{"@value":"Elkjaer"}],"schema:givenName":[{"@value":"Maria L"}]},{"@type":"schema:Person","schema:affiliation":[{"@type":"schema:Organization","schema:name":[{"@value":"University of Veterinary Medicine Vienna"}],"skos:exactMatch":["https:\/\/ror.org\/01w6qp003"]}],"schema:familyName":[{"@value":"Erben"}],"schema:givenName":[{"@value":"Reinhold G"}],"skos:exactMatch":[{"@type":"ids:orcid","@value":"0000-0003-0801-6958"}]}],"schema:numberOfPages":["17"]}
