    <ns1:description language="de">Diese wissenschaftliche Arbeit hatte zum Ziel, ZusammenhÃ¤nge zwischen Proteinexpression des ZelladhÃ¤sionsmolekÃ¼l CD146 und dem Wachstum von caninen oralen malignen Melanomzellen aufzuzeigen. Nach Schmid et al. (2019) wird das Protein CD146 auf der ZelloberflÃ¤che und intrazellulÃ¤r von caninen Melanomzellen und Metastasen exprimiert, fehlt jedoch bei benignen Muttermalen (Lei et al., 2015). Diese Merkmale lassen vermuten, dass die Expression von CD146 eine Rolle bezÃ¼glich InvasivitÃ¤t, Progression und Prognose von caninen oralen malignen Melanomen spielen kÃ¶nnte. Aufgrund der AggressivitÃ¤t und hohen Metastasierungs-und MortalitÃ¤tsrate, die diese Art von Tumoren bei Hunden aufweisen, gilt es nach Ursachen und vor allem weiteren TherapiemÃ¶glichkeiten zu forschen. Aus verschiedenen, histopathologisch charakterisierten caninen Melanomzelllinien wurde jene mit der hÃ¶chsten CD146-Expression ermittelt und fÃ¼r die weiteren Versuche verwendet. FÃ¼r die Herabregulierung von CD146 wurden verschiedene siRNAs sowie unterschiedliche Zeitpunkte fÃ¼r die Transfektion mit siRNA mittels Zeitkurve getestet. Das weitere Vorgehen basierte auf der DurchfÃ¼hrung von Western Blots zur Bestimmung der optimalen siRNA- Konzentration, der optimalen Inkubationszeit und Einfluss der CD146-Expression auf die Proliferation von caninen primÃ¤ren Melanomzellen. Die Ergebnisse zeigten, dass die Zelllinie cRGO1 die hÃ¶chste CD146-Expression aufwies. Die Inhibition der Expression wurde mit allen verwendeten siRNAs #s530434,35 und 36 in den jeweiligen Konzentrationen 10, 25 und 50 nM erzielt. Die weiteren Versuche wurden mit siRNA #s530434 in der Konzentration 10 nM vorgenommen. Achtundvierzig und 72 h nach Transfektion mit siRNA #34 wurde die Herabregulierung der Expression von CD146 am deutlichsten. BezÃ¼glich dem Proliferationsverhalten caniner oraler maligner Melanomzellen nach siRNA- vermittelter Herabregulierung von CD146 konnte mittels AuszÃ¤hlung der Gesamtzellzahl sowie Lebendzellzahl nach 0, 24, 48 und 72h gezeigt werden, dass die Proliferation von cRGO1 durch CD146-Inhibition signifikant gehemmt wurde. CD146-inhibierte cRGO1-Zellen proliferierten nach 48h und 72h signifikant weniger als mit Kontroll-siRNA transfizierte cRGO1. Zusammenfassend kann man sagen, dass die Proliferation von cRGO1-Zellen unter anderem von CD146 abhÃ¤ngig ist und durch Inhibition dieses Proteins mittels siRNA das Wachstumsverhalten von caninen oralen malignen Melanomzellen vermutlich generell beeinflusst werden kann. Diese Erkenntnis kÃ¶nnte sich als wertvoll in Hinblick auf zukÃ¼nftige TherapiemÃ¶glichkeiten beim caninen oralen malignen Melanom erweisen.</ns1:description>
    <ns1:description language="en">The aim of the thesis was to assess correlations between expression of the cell adhesion protein CD146 and proliferation of primary canine oral malignant melanoma cells. According to Schmid et al. (2019) the expression of CD146 occurs on the surface of and within canine melanoma cells and metastases. These features presume that CD146 expression may serve as diagnostic marker with respect to invasiveness, progression and prognosis of canine oral malignant melanoma. Due to the aggressiveness and high metastatic potential of these canine neoplasms, further research is needed regarding their etiology and innovative treatment options. Different canine melanoma cell lines were tested for CD146 expression. The cell line with highest expression rate was used in further experiments. The downregulation of CD146 was executed with different siRNAs and also at different points to generate a time curve revealing the optimum siRNA incubation time. Western Blots were used to identify the ideal siRNA concentrations and incubation time and also to evaluate the impact of CD146 expression on proliferation of canine primary melanoma cells. The results showed that the highest expression of CD146 was found in cell line cRGO1. The inhibition of expression was achieved by all used siRNAs (#s530434, 35 and 36) at all concentrations assessed (10, 25 and 50 nM). SiRNA #s530434 at a concentration of 10 nM was used in further experiments. Inhibition of CD146 expression by this siRNA was most effective 48 and 72 hours post infection. Regarding the proliferation behavior of canine oral malignant melanoma cells after CD146 downregulation by siRNA, total and living cell counting revealed that the proliferation of cRGO1 cells was blocked by CD146-inhibition. In conclusion, proliferation of cRGO1 cells depended on CD146 expression, suggesting that canine oral malignant melanoma cells may need CD146 for their proliferation. Provided that this assumption can be confirmed, inhibition of CD146 may be an interesting treatment strategy for canine oral malignant melanoma in the future.</ns1:description>
