    <ns1:description language="de">Das Porcine Reproductive and Respiratory Syndrome Virus, kurz PRRSV, gehÃ¶rt zur Familie der Arteriviridae, liegt einzelstrÃ¤ngig sowie in positiver PolaritÃ¤t vor. Es ist der Verursacher einer der wichtigsten Erkrankungen in der Schweinemedizin, welche jedes Jahr hohe wirtschaftliche Verluste verursacht. Die VerfÃ¼gbarkeit infektiÃ¶ser cDNA-Klone bietet die MÃ¶glichkeit zur Analyse, und Modifizierung viraler Genome auf molekularer Ebene und hat die Erforschung der Virusreplikation, Pathogenese und der Entwicklung von neuen Impfstoffen stark unterstÃ¼tzt. Das Ziel dieser Arbeit war es, einen molekularen cDNA-Klon des Feldisolates Aut 14-440 zu konstruieren, da dieser Besonderheiten im Genom sowie in der von ihm ausgelÃ¶ster Klinik aufweist. Anders als bei frÃ¼heren VersuchsansÃ¤tzen, bei denen durch zahlreiche Klonierungsschritte hÃ¤ufig Fehler in die Sequenz gelangen konnten, wurde durch die Kombination von neuen, leistungsfÃ¤higen Enzymen fÃ¼r die PCR und der âGibson assemblyâ Technologie ein hÃ¶herer Grad an SequenzstabilitÃ¤t bei den im Rahmen dieser Arbeit durchgefÃ¼hrten Versuchen erreicht. Die wichtigsten Klonierungsschritte waren die Ermittlung des korrekten 5â Endes, die Ermittlung des korrekten 3â Endes und die Klonierung mÃ¶glichst langer zusammenhÃ¤ngender cDNA Fragmente. Der finale Klon, pAX3, wurde in empfÃ¤ngliche Zellen mittels Elektroporation eingebracht und nach einer Inkubationszeit von 48 Stunden wurden diese Zellen fixiert, permeabilisiert und mit einem Cy5 konjugiertem AntikÃ¶rper gegen das N-Protein inkubiert. Ohne Virusreplikation in der Zelle wÃ¤re auch kein N-Protein nachweisbar. Das bedeutet im Umkehrschluss, dass eine im Fluoreszenzmikroskop erkennbare rote FÃ¤rbung einzelner Zellen ein guter Hinweis auf stattgefundene Virusreplikation ist. Dies war, zusammen mit der Verteilung des Antigens im Zytoplasma, ein Beweis dafÃ¼r, dass pAX3 replikationsfÃ¤hig ist.</ns1:description>
    <ns1:description language="en">The Porcine Reproductive and Respiratory Syndrome Virus, PRRSV for short, belongs to the family Arteriviridae, is single-stranded and has a positive polarity. It is the cause of one of the most important diseases in pig medicine, which causes high economic losses every year. The availability of infectious cDNA clones provides the potential for analysis and modification of viral genomes at the molecular level and has strongly supported the study of virus replication, pathogenesis, and the development of new vaccines. The aim of this work was to construct a molecular cDNA clone of the field isolate Aut 14-440, as it exhibits peculiarities in the genome as well as in the clinical symptoms that it triggered. Unlike previous approaches, where multiple cloning steps often allowed for errors in the sequence, the combination of new, high-performance enzymes for PCR and the Gibson assembly technology resulted in a higher level of sequence stability in the experiments performed in this work. The most important cloning steps were the determination of the correct 5 'end, the determination of the correct 3' end and the cloning of contiguous cDNA fragments as long as possible. The final clone, pAX3, was electroporated into susceptible cells and after 48 hours of incubation, these cells were fixed, permeabilized and incubated with a Cy5-conjugated antibody against the N-protein. Without virus replication in the cell, no N-protein would be detectable. This implies, conversely, that a red staining of individual cells recognizable in the fluorescence microscope is a good indication of the virus replication that has taken place. This, together with the distribution of the antigen in the cytoplasm, proved that pAX3 is replicable.</ns1:description>
