    <ns1:description language="de">Lamin A wird als PrÃ¤lamin A translatiert und anschlieÃend carboxyterminal am -CAAX Motiv posttranslational modifiziert. Die -CAAX-Box wird farnesyliert, endoproteolytisch gespalten und carboxymethyliert. Danach werden die letzten 15 AminosÃ¤uren, inklusive farnesylierter -CAAX-Box, proteolytisch abgeschnitten. Beim Hutchinson-Gilford-Progerie-Syndrom bleibt das mutierte Lamin A Protein dauerhaft farnesyliert, weshalb die Patient*innen den PhÃ¤notyp des vorzeitigen Alterns tragen. Ein bereits zur Behandlung zugelassener Farnesyltransferaseinhibitor, Lonafarnib, soll die Farnesylierung verhindern und dadurch zu einer Verbesserung des Krankheitsbildes fÃ¼hren. Eine Lonafarnib Behandlung induziert eine PrÃ¤lamin A Akkumulierung in mesenchymalen Stromazellen. Der Effekt der Behandlung ist jedoch begrenzt, was auf eine alternative Prenylierung durch Geranylgeranyl zurÃ¼ckzufÃ¼hren sein kÃ¶nnte. Farnesyl und Geranylgeranyl sind Produkte des Mevalontweges. Statine und Bisphosphonate inhibieren unterschiedliche Schritte genau dieses Syntheseweges. Statine greifen zu einem frÃ¼hen Zeitpunkt ein, indem sie die 3-Hydroxy-3-Methylglutaryl-Coenzym-A-Reduktase inhibieren. Bisphosphonate hingegen inhibieren die Farnesylpyrophosphatsynthase und Geranylgeranylpyrophosphatsynthase. Eine Triple-Medikament Therapie bestehend aus Lonafarnib (Farnesyltransferaseinhibitor), Zoledronate (Bisphosphonat) und Pravastatin (Statin) sollte bei Hutchinson-Gilford-Progerie-Syndrom Patient*innen Besserung gegenÃ¼ber der Lonafarnib Monotherapie bringen, welche jedoch aus bis jetzt nicht bekannten GrÃ¼nden nicht eintrat. In dieser Arbeit wurde mittels Immunfluoreszenzmikroskopie und Western Blot der Einfluss von Statinen, Bisphosphonaten und Lonafarnib in murinen mesenchymalen Stromazellen auf die Prozessierung von Lamin A, die allgemeine zellulÃ¤re Farnesylierung und die osteogene Differenzierung untersucht. Mit einer alternativen Kombinationsbehandlung bestehend aus Lonafarnib und einem lipophilen Bisphophonat konnte eine verbesserte Prenylierungsinhibition geschaffen werden. Gleichzeitig konnte eine neue Perspektive zur Bewertung von Prenylierungsinhibitionen erprobt und ein vermeintlicher Nebeneffekt von Farnesyltransferaseinhibitoren identifiziert werden.</ns1:description>
    <ns1:description language="en">Lamin A is translated as prelamin A and subsequently post-translationally modified. The modification occurs carboxy-terminal at a so-called CAAX box. The CAAX-box becomes farnesylated, endoproteolytically cleaved, and carboxymethylated. In a final step, the last 15 amino acids, including the farnesylated CAAX-box, are proteolytically cleaved off. In Hutchinson-Gilford progeria syndrome, the mutated lamin A protein remains permanently farnesylated, which causes the phenotype of premature aging. A farnesyltransferase inhibitor already approved for treatment, lonafarnib, prevents farnesylation, thus leading to an improved disease pattern. Lonafarnib treatment induces prelamin A accumulation in mesenchymal stromal cells. However, the effect of the treatment is limited, which may be due to alternative prenylation by geranylgeranyl. Farnesyl and geranylgeranyl are products of the mevalonate pathway, which is inhibited by statins and bisphosphonates at different steps. Statins intervene at an early stage by inhibiting the 3-hydroxy-3-methylglutaryl coenzyme A reductase. Bisphosphonates, on the other hand, inhibit the farnesyl pyrophosphate synthase and the geranylgeranyl pyrophosphate synthase. A triple-drug therapy consisting of lonafarnib (farnesyltransferase inhibitor), zoledronate (bisphosphonate), and pravastatin (statin) was expected to improve treatment outcomes compared to the lonafarnib monotherapy in Hutchinson-Gilford progeria syndrome patients. However, no improvement was observed due to reasons yet unknown. In this study the influence of statins, bisphosphonates and lonafarnib on the processing of lamin A, on the cellular farnesylation in general, and on the osteogenic differentiation was investigated in murine stromal cells. Using an alternative combination treatment consisting of lonafarnib and a lipophilic bisphosphonate demonstrated an improved prenylation inhibition. Furthermore, a new perspective to evaluate prenylation inhibition was tested and a potential side effect of farnesyltransferase inhibitors was identified.</ns1:description>
